Anil Potti faked cancer research at Duke — patients were treated based on his fraud

Duke's star cancer researcher fabricated gene-expression data for years. His work sent three clinical trials treating real patients — all based on fraud.

What happened

Anil Potti was a rising star at Duke University — a physician and cancer researcher who claimed to have developed gene-expression signatures that could predict how a patient would respond to specific chemotherapy drugs. His work, published in top journals including Nature Medicine and the New England Journal of Medicine, promised personalized cancer treatment based on genomic analysis. Duke launched three clinical trials treating real cancer patients based on Potti's research.

Doubts emerged in 2007 when statisticians at MD Anderson found Potti's data looked too clean to be real. In 2010 The Cancer Letter revealed Potti had fabricated his Rhodes Scholar claim. Duke suspended the trials; a medical student's 2008 memo — which Duke dismissed — was re-examined. The FDA found one trial lacked a required Investigational New Drug application. Co-researcher Nevins called it 'manipulated data' that could not be chance.

The Office of Research Integrity officially found Potti engaged in research misconduct in November 2015. Eleven of his papers were retracted; Duke's own expert testified roughly 27 of 40 examined papers would need retraction. Duke settled malpractice claims with cancer patients. Potti lost funding, was fired from his next job, and was banned from receiving federal research funds until 2020. The scandal prompted an Institute of Medicine report on oversight of genomic clinical trials.

The lesson

Biomedical data fabrication is not just academic fraud — when results guide clinical trials, fake data means real patients get wrong treatment. One researcher can corrupt an entire institution's work.

Aftermath

The Anil Potti scandal became a landmark case in research integrity. The Institute of Medicine's 2012 report set new standards for validating genomic signatures before clinical use. Duke overhauled its conflict-of-interest practices. The case taught that even prestigious institutions can be fooled by fabricated data, and that the cost of trusting attractive results can be measured in patient harm. Potti returned to medicine, but his research career was destroyed.
